In Coming to America, Prince Akeem, played by Eddie Murphy, is determined to find a wife who loves him for who he is, rather than simply because of his royal status. Growing tired of his arranged marriage, Akeem convinces his father, King Jaffe Joffer (James Earl Jones), to give him 40 days to travel to America and find a suitable bride. Akeem's loyal servant Semmi (Arsenio Hall) accompanies him on this journey.

Upon arriving in Queens, New York, Akeem and Semmi decide to live humbly and under the radar, taking on jobs at a local fast-food restaurant called McDowell's. Akeem disguises his royal identity and assumes the name "Prince" while Semmi poses as his chauffeur.

During their stay in Queens, Akeem encounters Lisa McDowell (Shari Headley), a beautiful and independent woman who works at her father Cleo McDowell's (John Amos) fast-food restaurant. Akeem is immediately drawn to Lisa's strong-willed personality, intelligence, and ambition, making her the ideal woman he had hoped to find.

To win Lisa's heart, Akeem must navigate both cultural differences and the disapproval of Lisa's boyfriend, Darryl (Eriq La Salle). As Akeem continues to court Lisa, he must also keep his true identity hidden from her and her family, fearing that the revelation of his royal heritage could hinder their genuine connection.

As the days go by, Akeem and Semmi experience various humorous and challenging situations in Queens. They encounter a variety of colorful characters, including Akeem's landlord and confidant, Mr. Randy Watson (Eddie Murphy), who he meets at a local barbershop.

Complications arise when Lisa discovers Akeem's true identity. Feeling betrayed, she rejects his advances, causing Akeem to question his journey and the worthiness of his pursuit. However, Akeem's genuine and selfless intentions eventually win Lisa and her family over, and they reconcile.

In a climactic finale, Akeem returns to Zamunda to face his father, who is furious with his decision to marry an American commoner. However, Akeem's display of true love and commitment convinces King Jaffe Joffer to bless their union, and Akeem and Lisa are married in a grand ceremony.
