Miles Morales, a teenager living in Brooklyn, is struggling to fit in at his new prestigious high school while dealing with the pressures of his overachieving policeman father, Jefferson, and his supportive, yet strict mother, Rio. One night, Miles wanders into an abandoned subway station and is bitten by a radioactive spider, gaining spider-like powers.
Soon after, Miles witnesses the battle between Spider-Man and the villainous Green Goblin at his universe's version of the Super Collider, a device created by the criminal mastermind, Wilson Fisk, to access parallel dimensions. Spider-Man tries to stop the collider, but is ultimately killed in the fight.
Following Spider-Man's death, Miles encounters the gravely injured Peter Parker, who is from another universe. Peter reveals the existence of the multiverse and suggests that the collider caused other Spider-Men from various dimensions to be displaced and stranded in Miles' universe. Peter agrees to train Miles to become the new Spider-Man so they can shut down the collider and send everyone back to their respective dimensions.
Together, Peter and Miles embark on a mission to gather the other displaced Spider-People, which includes the cool and confident Gwen Stacy, also known as Spider-Woman; the middle-aged Peter B. Parker from a universe where his life took a dark turn; Spider-Man Noir, a gritty detective from a 1930s universe; Peni Parker, a young girl from a future Japan piloting a mechanized spider suit; and her spider mascot, Spider-Ham, a cartoon pig with spider powers.
As they face their own personal struggles and adjust to their new reality, the team of Spider-People must work together to stop Wilson Fisk's plans and restore the balance of the multiverse. Along the way, Miles must also confront his doubts and fears as he grows into his role as Spider-Man and realizes that he has the power to save his universe and become a hero in his own right.
In a climactic battle, the team manages to shut down the collider, but not before sacrificing some of their own, including Peter B. Parker. Miles, now confident in his abilities, takes up the mantle as the one and only Spider-Man of his universe, protecting his city from Fisk and other threats.
In the end, Miles embraces his role as Spider-Man, inspiring others and becoming a symbol of hope. With the collider repaired, the displaced Spider-People bid their farewells and return to their respective universes, but not before leaving a lasting impact on Miles and his newfound determination to protect his world.
