In the movie Sister Act, the Reno singer, Deloris Van Cartier (played by Whoopi Goldberg), witnesses a mob murder committed by her boyfriend, Vince LaRocca (played by Harvey Keitel). Fearing for her life, Deloris goes to the police for protection. The police come up with a plan to hide her in a convent, believing that the mob hitmen would never suspect to find her there.

At the St. Katherine's Convent in San Francisco, Deloris is placed under the care of the strict Mother Superior (played by Maggie Smith). However, the Mother Superior harbors reservations about Deloris' wild, irreverent personality and doubts her commitment to the religious life. Mother Superior assigns Deloris the role of Sister Mary Clarence and restricts her interactions with the other nuns.

Deloris initially struggles with the strict and disciplined lifestyle at the convent. She finds it difficult to conform to the rules while still maintaining her own vibrant personality. However, she gradually forms friendships with some of the nuns, particularly Sister Mary Patrick (played by Kathy Najimy) and Sister Mary Lazarus (played by Mary Wickes).

One day, while attending the convent's failing choir practice, Deloris realizes that the choir lacks enthusiasm and musical talent. To bring life into the choir and make it more engaging, she volunteers to take over as the choir director. Deloris introduces gospel music and transforms the choir into a dynamic, energetic group. As a result, the choir becomes popular within the community and brings in more people to attend the church.

Deloris begins involving the nuns in various community projects, such as cleaning up the neighborhood and assisting the less fortunate. These actions not only help the community but also bring Deloris closer to the other nuns, gaining their trust and respect.

Later, the choir's newfound success catches the attention of a television producer, resulting in an interview opportunity for Deloris. During the television interview, Deloris inadvertently reveals her identity while talking about her past life as a Reno singer. This revelation is witnessed by Vince and his mob associates.

As the mob closes in on Deloris at the convent, she and the nuns work together to protect each other. Deloris helps the nuns not only with their musical talents but also with their newfound strength and self-confidence. The quirky group of nuns successfully manages to outwit the mob and deliver them into the hands of the police.

In the end, Deloris and the nuns give a show-stopping performance at a grand concert, showcasing their new musical abilities. Deloris's talent, combined with the unity and faith of the nuns, brings joy and rejuvenation to the convent and community. Deloris ultimately finds her true calling in music and selflessly decides to stay at the convent to continue helping the nuns and spreading love and joy through her music.
